P21

P21 protein sits in normal cells in the form of tetrameric complex which formed with cell cyclin, cyclin dependent kinase(CDK), and proliferating cell nucleoantigen as well as proliferation cellular nuclear antigen. In addition to inhibit phosphorylating of RB protein, and to cause the cell cycle to stall in the G1 phase. Ras cancer gene activation can be mutated by the point mutations of the 12th, 13th, 59th, 61th genetic code, or by gene amplification. Because of the activation of Ras cancer gene, the P21 protein will occur abnormal expression. P21 protein has the higher conservative in term of evolution and has a G-protein function.The GTP enzyme of mutated P21 protein decreases the activity, then it can not carry on the hydrolysis from GTP to GDP quickly.Thus, the cell produces a continuous proliferation, which eventually leads to cell cancer. The results show that P21-Ras is normal and atypical proliferating in the general hyperplasia of the breast catheter epithelial and the expression in the invasive ductal carcinoma is increased. The positive expression rate in lung cancer can reach 72.94%.
