Multiplex Immunofluorescence (mIF) is an advanced technique widely used in pathology and biomedical research to simultaneously detect and visualize multiple biomarkers within a single tissue section. It enables detailed analysis of protein expression, interactions, and spatial distribution within the tissue microenvironment.
Chromogenic multiplex IHC (mIHC) is a simpler alternative suitable for routine use; however, it is typically limited to the detection of up to four markers per slide. Moreover, chromogenic methods often struggle to resolve co-localization of markers—particularly those expressed on membranes or within nuclei—whereas fluorescence-based approaches excel in this regard.
Historically, the use of multiple antibodies, whether in a mixed protocol or sequentially, has been constrained by the specificity of primary antibodies and the risk of cross-reactivity leading to false positives. As a result, most studies relied on single or limited multi-parametric (e.g., dual or triple) staining approaches, interpreted manually by researchers.
Today, mIF is considered an essential tool in tumor microenvironment (TME) research, providing high-dimensional insights critical for understanding complex cellular interactions in cancer biology.
